在详细分析TNF结构及前人有关TNF结构与功能关系研究的基础上,应用PCR技术构建了一种新型人肿瘤坏死因子(TNF)分子的编码基因,并将该编码基因插入表达质粒,转化大肠杆菌,通过温度诱导获得了高表达.活性测定的结果表明,新型人TNF对L929细胞的细胞毒活性较原型人TNF升高了103数量级.
On the basis of analysis of TNF structure and the relationship between structure and function,a novel TNF coding sequence was synthesized by PCR technique and inserted into an expression plasmid. By temperature induction the transformed E. Coli with the novel TNF expression plasmid produced high yield of novel TNF, whose cytotoxic activity to L929 cell was 103 higher than recombinant human TNF.
