    Abstract:

    Protein C-terminal sequence analysis is an important technique in protein chemistry and molecular biology. (iso)Thiocyanate approach appears to be the prevailing method since it was first described by Schlack and Kumpf in 1926. The chemical mechanism, the advances of (iso)thiocyanate approach and the preparation of amino acid thiohydantoins are reviewed and discussed in detail.
