利用细胞裂解液充分溶解细胞蛋白质是成功进行蛋白质组分析的先决条件.尝试利用三步提取法,即以三种溶解性能不同的裂解液分步提取细胞中的蛋白质组,并分别进行二维聚丙烯酰胺凝胶电泳(two-dimensional polyacrylamide gel electrophoresis, 2-D PAGE)分离.通过对2-D PAGE蛋白质图谱的比较,发现其与常规方法相比,具有蛋白质提取率高、双向电泳(two-dimensional electrophoresis,2-DE)分辨率高等优点.
A three-step sequential protein extraction method was applied in proteomics analysis: using three kinds of solutions with differential solubility to replace one kind of solution. The method increases the efficiency of extraction and separation of the proteins compared with method of one solution.
