Some ways are introduced to enhance the cloning efficiency of PCR amplification produts. The purification of PCR products，the speciality of PCR amplification, the remainder of Tag polymerase, the 3′-end projection of PCR products and the blunt end ligation are the main factors to affect the cloning efficiency.
