Genomic DNA extracted from the posterior silk glands of two silkworm varieties,C108 and Dazao were digested with PstⅠ and ligated with synthetic adapter.Selective amplification of DNA fragments （SADF） was performed using suitable primers which annealed to adapter regions.Agarose gel electrophoresis shown that DNA polymorphic fragments were appeared for both C108 and Dazao.
