Abstract:Various sequence blocks. due to their specific primary sequence box, and/or a curved helical DNA conformation. and/or a non-β-helical DNA structure, can be organized in the corresponding higher order structures and behave as functional sequence domains recognized and bound by specific proteins. They may be defined as “code domains”. Code domains are genetically instructive for specific molecular interactions or processes, important not only in nucleus during interphase and during cell division but also in diferential gene expression during development and differentiation.

Abstract:The studies in this lab on the enzyme system of respiratory chain were reviewed briefly. Two inhibitors which affect simultaneously on three Q-related enzymes of complex Ⅰ.Ⅱ and N were synthesized. They could be an useful tools in the studies of ubiquinone reactions in respiratory chain. The direct interaction between TTFA inhibitive site of QH→QH2 of complex Ⅱ and cytochrome b562 of complex Ⅲ was found. This kind of interactions may playsome roles in modulating the electron transfer from complex Ⅱ to complex Ⅲ. The maximum reconstitutive activity of lipid-deplited succinate-cytochrome c reductase with mixed phospholipids PC:CL:PE=2:2:1 were obtained. this means not only the lipid but also thier composition were important for regulating the enzyme activiy of respiratory chain.

Abstract:Studies on ribosomal RNA topography play an important role toward elucidation of the functional roles of rRNAs in protein synthesis.RNA N-glycosidases are a group of ribosome-inactivating proteins which inactivate ribosomes by hydrolyzing the N-C glycosidic bond of a special adenylic acid in rRNA and releasing the adenine. Ricin A-chain is the first determined RNA Nglycosidase which has been studied in the greatest detail. UP to the present. 25 glycosidases have been characterized. The action site of RNA N-glycosidases lies in the a-sarcin domain of 28S rRNA. Treatment of RNA N-glycosidases induces conformational change of ribosomes and leads to the inactivation.

Abstract:As a family of multifunctional cell-proliferation regulating factor. TGF-β has great potential in clinical application. Both naturaly secreted and recombinantly expressed TGF-ss are existed in the inactive form of latent complex. Activation of latent TGF-β complex is an important pathway of modulating the biological function of TGF-β. This review concerns on the molecular structure of both natural and recombinant latent TGF-β complexes, and their possible activation mechanisms under physiological conditions.

Abstract:Human factor Ⅷ is an important cofactor in the intrinsic blood coagulation. Hemophilia A is the most common severe inherited bleeding disease due to the deficiency or abnormality of factor Ⅷ. Factor Ⅷ gene has been successfully cloned and expressed in eukaryotic cells that promotes the studies on the gene mutations of factor Ⅷ widely and thoroughly. This article introduces the recent progress about this field. and new techniques used in researches. The study on gene abnormalities of factor Ⅷ can be regarded as an excellent example both in depth and width for researches of the congenital diseases.

Abstract:Results of a series of studies by this research group on the functional roles of the callosal connections in primary visual cortices were reviewed. Based on these, a topographical projection model of the vertical retinal bilaterally projecting strip was proposed. which not only agrees well with experimental results but also explains the necessity of the corpus callosum: signals conveyed by these fibers compansate information loss in the cortex due to the existence of retinal bilaterally projecting strip.

Abstract:Recent studies of cell biophysics have been introduced about cell ultrastructure effects of physical factors on living cells, cellular motilities, ion channels and cellular signalling. Some progress in methodology for studying living cells are also reviewed. It has been emphasized that the main purpose of cell biophysics is to understand the nature of the living cell in order to explain why and how the cell is alive. Accordingly, some questions and problems on principles and methodology have been discussed in this article as well.

Abstract:Adriamycin (ADM) is a widely used anticancer drug, but the chronic cardiotoxicity severely limits the use of it in the treatment of neoplastic disease. The experimental results obtained from F1F0-ATPase activity assay. 31P-NMR spectra measurement. fluorescent probe NBDPE detection. packing and fluidity of membrane lipids and intrinsic fluorescence measurements can be summarized as follows: ADM induces the phase transition of mitochondrial membrane lipids at first. as a consequence affecting on the lipid packing and fluidity of the lipid molecules and then influencing the conformation of the F1F0-ATPase and finally resuiting in the decreasing of the enzymatic activity. And Mg2+ can protect all the effects induced by ADM thus reducing the harmful effect of ADM.

Abstract:Vacuum ultraviolet circular dichroism (VUV-CD) spectra of proteins in solution have been measured using synchrotron radiation vacuum ultraviolet circular dichroism spectrometer and a special cell. The measurement wavelength is down to 175nm. A new calculation method has been applied for calculating the content of five kinds of secondary structures of proteins. Their results are coincident with that from X-ray diffraction method. In order to get good VUV-CD spectra, several important factors have been discussed. The experiments show that so far. VUV-CD analysis is one of the favorable method for secondary structure studies of proteins.

Abstract:The full-length intronless firefly luciferase gene and its deletion mutant which lacked 48 nucleotides from the initiation codon (ATG) were inserted into the high expression secretion vector. pIN-Ⅲ-ompA3. and introducedinto E. coli cells in which high level and no luciferase activity were detected, respectively.This result shows that the first sixteen N-terminal amino acids of firefly luciferase involve in catalytic activity.

Abstract:The extracellular catechol 1, 2-dioxygenase was immobilized. The apparent activity of the immobilized extracellular catechol 1, 2-dioxygenase was high. and range of use was extended. The pH-activity profile was altered by immobilization, and optimal PH from 6.0-9.0 was observed. Immobilization was shown to increase the thermal stability of the enzyme.The pureness and concentration of biotrans-formation of catechol to cis, cis-muconic acid were high. The product was easily separatedfrom enzyme. The immobilized method of extracellular catechol 1,2-dioxygenase was novel and simple. The results presented show that the immobilization of extracellular catechol 1,2-dioxygenase offers an attractive means for the production of cis, cis-muconic acid.

Abstract:The nucleotide sequence of bovine liver tRNAIle was determined by both the partial hydrolyzation in water and the Donis-keller method.The tRNAIle was consisted of 77 nucleotides and relatively rich in GC base pairs. The acceptor stem of the tRNAIle was characteristic of G5 . G69 mismatch. Forthermore, the cloverleaf structure of tRNAIle was showed according to the Holley mode as well as the amount of free energy in the each of stems versus loops.

Abstract:The strength of radiation of 45Ca2+ in sarcoplasmic reticulum vesicle (SR) and mitochondria (Mit) was measured by liguid scintillation counting method. Comparing the effects of ATP - MgCl2 . SOD and verapamil on the 45Ca2+ concentration in SR and Mit which prepared from isolated ischemia reperfused rat hearts. The result showed that the 45Ca2+ cpm of SR of the three groups were higher than that of control, while the 45Ca2+ cpm of Mit of the three groups were lower than that of control. These results suggested that the three kinds of reagent could protect reperfusion injury of heart cell through enhance 45Ca2+ storage in SR and inhibitting the 45Ca2+ accumulation in Mit.

Abstract:A new kind of diazafluorenone Schiff base amphiphiles were synthesized. Bilayer membrane formed with these compounds possess good stability and oscillation properties. When 0.1mol/L AgNO3 presented in bathing solution and an electrical field was applied on this system, a maximum value of the current. 4.0 PA, was obtained. A possible application in the development is indicated.

Abstract:The culture solution of recombinant strains of lysozyme was treated by ultrasonic wave, purified by DE52 chromatography and CM52 chromatography. a polyacrylamide gel eletrophoresis pure T7 lysozyme of Mr 17000 was obtained. the optimal react PH is 8.0, 21% of total enzyme activity lost at 37℃ in 5 min.

Abstract:A rapid and sensitive method of isoforms of CK (EC 2.7.3.2) -MM in human serum by chromatofocusing was reported. The assay system involved Mono P (HR5/20) column. fast protein liquid chromatography (FPLC) system and post column reactor. The result of CK-MM isoforms was obtained within 40 min after serum application to equilibrated column.The lower limit of sensitivity for CK activity is 8U/L and the linear capacity is 17-2085U/L, with CV values of 2.1%. 2.7% and 11.0% for MM1. MM2 and MM3 respectively.
