Abstract:Interleukin 6 (IL-6) is a multifunctional cytokine,whose biological effects mainly depend on the structure and function of IL-6 receptor. IL-6R is composed of two polypeptide chains,a ligand-binding receptor gp80(IL-6Rα),and a nonbinding signal transducer gp130 (IL-6Rβ).The two subunits share the function and cooperate with each other,and form high affinity IL-6R after gp80 chain binding IL-6. The haemopoietic domain in IL-6Rα chain is capable for binding IL-6. However,gp130 is shared by several cytokines as their common signal transducer. Cytoplasmic portion of gp130 has some conserved amino acids which are related to actieating some tyrosine kinases .The homodimerization of gp130 induced by IL-6+IL-6R complex can activate a series of kinases and transcription factor and finally leads to related genes expression.

Abstract:Human erythropoietin receptor (hEPOR) is a transmembrane protein which located in the surface of the relatively mature erythroid progenitor cells. It can promote the viability,proliferation and differentiation of the cells by specifically binding to erythropoietin(EPO),in which process the hEPOR itself and some other proteins will be phosphorylated. There are some conserved domains in hEPOR which is adapted to its function,The amino acid sequence of the human EPOR is 82% identical to that of the mouse protein. The EPOR can be constitutively activated by a single R129C mutation or binding to a special protein. It is also closely related to many blood diseases such as erythroleukemia.

Abstract:Steroid hormone receptor superfamily is a kind of gene trans-acting factors and modulates both RNA pol -directedr RNA transcription and RNA pol Ⅱ-directed transcription. The melocular pathway of RNA pol Ⅱ-directed transcription regulated by the superfamily includes the binding of ligand, dissociation of relative proteins, receptor phosphorylation,homo/hetero-dimerization,nuclear localization and interaction with positive or negative hormone response elements and related transcriptional proteins. As a result,the specific target gene is activated or repressed. Now more studies focus on the actieating mechanism of some classic receptors and orphan receptors with or without ligand,as well as their regulations on specific gene transcription.

Abstract:It is well known that zinc is an essential element for life. It takes part in a variety of important metabolisms in biological systems in the form of zinc containing metalloen-zymes. In the past decade, it found that zinc is also involved in the gene transcription,duplication and protein synthesis processes as a regulator and mediator. Till now,a number of zinc proteins have been charcterized such as zinc finger,zinc twist,zinc ribbon and zinc cluster. Among them the zinc-metallothionein, Zn7-MT is one of the well known zinc-cluster protein which is suggested palying a crucial role in the zinc homeostasis. It is very possible that zinc occupies the central part in controlling the growth and development of organisrns by means of successive regulation of MT and zinc finger proteins. In addition,it was found that some of regulation protein binding to oncogene and human immunedeficiency virus also contains zinc finger amino acid sequence,that will open a new field to fight against cancer and aids diseases.

Abstract:MK is a new identified cytokine, which belongs to the heparin binding family. MK molecule is a small polypeptide whose gene expresses only in mid-gestation embryo and adult kidney.The expression of MK gene has also been shown in various carcinoma cells. MK can promote the growth and differentiation of normal cells,particularly the development of nerve cells.MK can also inhibit the growth of several kinds of carcinoma cells.The reason why MK gene is expressed in adult kidney has not been elucidated.

Abstract:Intracellular signalling(or signal transduction)has been one of the most active research areas in biological science since early 80's. One of the major objectives of this research field is to understand the mechanism of an organism's response to stimuli from the environment.Recent research has focused on the biochemical and physiological changes in the cell following the initial transmembrane signalling event.The recent progress in this research area is briefly summarized with special emphasis placed on the newly proposed models for various intracellular signalling pathways in animal cells.

Abstract:The recent results in the research of gene therapy have been encouraging,in spite of numerous problems still remaining to be solved.The basic studies on gene therapy and the study on the techniques of the gene introduction are reviewed.The latter is one of the most important topics in basic studies, including retrovirus vectors,DNA virus vectors as well as non-viral techniques.The recent advances in the establishment of the experimental models are also described.Then the recent advances in the gene therapy of hereditary diseases, malignant tumors and other diseases are briefly discussed, especially emphasizing the strategies of the gene therapy of AIDS and cardiovascular diseases.

Abstract:The various methods to make supported bilayer systems on solid substrates are straight foreward.Comparing with liposomes and other systems,these systems have the advantages of better reproducibility,tighter controls over their physicochemical properties.Since membrane proteins can be incorporated into these supported bilayers, they are ideal model systems in the studies of biomembranes. Along with the improvements in the investigation methods,studies on these membranous systems are progressing into deeper details and there have appeared more applications of these model systems in biological and medical researches. The producing and studying methods of surpported bilayer systems on solid substrate are reviewed,and some examples of successful applications of these systems in membrane biophysical chemistry are attempted to give.

Abstract:Cartilage angiogenesis inhibiting factor(CAIF)was purified from cartilage by guanidine hydrochloride extraction,acetone fractional precipitation, ultrafiltration,column chromatography.With SDS-PAGF,it showed a single band of CAIF with molecular weight of 27 700.By the methods of [ 3H]-TdR incorporation and the determination of mortality of cells,the effect of CAIF on the DNA synthesis and cytotoxicity of all cell lines were investigated and the inhibitory effect of CAIF on angiogenesis was measured by the experiment of chorioallantoic membrane of chicken embryos. The results showed that CAIF inhibited the endothelial cell strongly and Hela cell slightly;but not QGY7703 cell,human skin fibroblast,and mice myeloid cell. It also inhibited the angiogenesis in the chorioallantoic membrane of chicken embryos significantly. All these results suggest that CAIF has the inhibitory effect on angiogenesis. It is a specific angiogenesis inhibiting factor,and may be a useful drug on tumor therapy in some day.

Abstract:The intensity of seeds luminescence induced by γ radiation and its decay speed both decrease gradually after the seed have been irradiated.The decay rule of the luminescence intensity follow nearly the second order reaction dynamics rule. The perseed luminescence induced by perdose correlate significantly with the seedling relative height,and the perseed or pergram seed luminescence at the soaking begining time correlate extremely significantly or significantly with the seedling relative height,these suggest that the higher radiosesitivity of the seed the higher intensity of seed luminescence induced by γ radiation. After germinating,the luminescence intensity of the irradiated seed is lower than that of the no-irradiated seed,this is because of the radianon inhibit the seedling growth.

Abstract:The effect of ganglioside GM3 on the metabolism of phosphoinositides of human monocytoid leukemia J6-2 cells has been studied using the incorporation of[ 32P]Pi and [ 3H]inositol into them. The results show that GM3 inhibited the incorporation of[ 32P] Pi and [ 3H] inositol into phosphatidylinositol (PI),stimulated the incoporation of those into phosphatidylinosito1-4,5-bisphosphate (PIP2) and [ 32P] Pi into phosphatidylcholine (PC), inhibited the incorporation of [ 3H] inositol into inositol phosphates (IP3,IP2, IP ),and of [ 32P] Pi into phosphatidic acid (PA) in a dose-dependent manner. These results suggest that GM3 inhibits the PI turnover of J6-2 cells.

Abstract:Kumming mice were chosen as experimental anminals.Under the conditions of actue and repeatitive hypoxia and normal,the indivival relative percentage of ganglioside GM1,GD1a,GD1b and GT in the brain were measured by the CS-9000 thin-layer-scanning photometer respectively. Comparing with the group A of blank control without exposure to hypoxia , group B of experimental control exposured to hypoxia only once, and group D of another experimental control fed for two days after acute and repeatitive exposure to hypoxia for 4 times,the content of sialic acid significantly decreased and that of GT markedly increased in the experimental animals group C acutely and repeatedly exposured to hypoxia for 4 times;while the contents of ganglioside were also different between group A and D significantly. The results indicate that ganglioside in the brain may be involved in the development of tolerance to hypoxia in animals exposured to acute and repeatitive hypoxia and the proceeding was not a fast respone which may be restored.

Abstract:The effects of DNA conformation and carrier DNA on the efficiency of electroporation are studied using the expression of erythropoietin cDNA in CHO cells as an indicator. It turns out that 250mg/L carrier DNA increases the expression level by 3 fold,and linearized plasmid DNA is more suitable than supercoiled DNA to obtain stable transfection by electroporation. The results suggest that it is essential to linearize plasmid DNA and cotransfect with apporporiate carrier DNA to obtain stable and high level expression of foreign genes by means of electroporation.

Abstract:The purification of recombinant human SCF(rh SCF)expressed in E. coli. By McAb affinity chromatography which prepared by combining an anti-SCF McAb with NCR-Sepharose 4B was performed. At optimum conditions,the purity of rhSCF obtained was higher than 95%.The biological activity also increased apparently.

Abstract:Human muscle specific enolase (hMSE) plays an important role in the diagnosis of acute myomuscle infarction and muscular diseases. A hMSE RIA is developed.An affinity coefficient of antiserum is 5.1×109. 125I-hMSE as the tracer is prepared using improved BHR method and its binding rate with antiserum(1∶1000) is up to 50.16%. The variable coefficients of intra and inter-assay are 8.6% and 13%, respectively. The recovery rate is 95%-105%. The standards range from 5 to 320μg/L and the minimum detectable value is 5μg/L. In the assay there is an optimal reaction condition: 0.1mol/L pH7.4 PBS(5mmol/L MgSO4,0.1% BSA, 0.1% Toween-20, 0.1% NaN3,0.5% NaCl);and incabation temperature and times: 37℃,0.5h and 4℃,0.5h (as rapid measurement) or 4℃ 24h. On the basis of 65 healthy control,the hMSE levels are 23.9±10.9μg/L (x±s).A positive value is higher than 57μg/L. The hMSE levels are signifcantly increased in the patients with both acute myocardial infarction and muscular diseases.

Abstract:The chlorophyll-protein complexes of thylakiods and those of various PSⅡ preparations were analyzed by a low ionic strength native“green gel”system after solubilization by a mixture of detergents,with very little release of free pigment.The subunit composition and absorption spectra of some chlorophyll-protein complexes were studied also.This new native green gel system is simple and rapid,with release of very little free pigment.

Abstract:The quantity change of the copies with mismatch bases in PCR products was analyzed.The general formulas of numbers of copies in different modes after certain cycles (n) were gained and the math relation between the minimum(Rn)of the non-base-mismatch copy proportion of PCR products and the efficiency cycle numbers (N),the copy chain length(H)catalyzed by Taq polymerase and the mismatch frequency(f)was concluded by way of substituting step by step:Rn=(1-Hf/2)N-1(1-Hf) .The formula has a guidance sense for preparing the gene segments used for the gene expression by PCR technique.

Abstract:When Cmnese Hamster V79 cells were incubated with H2O2 of different concentration for 24h and 48h,different degree damages in cells have been found.The activity of SOD decreased and lipid peroxides contents increased in cultured cells.It is also shown that the activies of metabolites,such as GPT,GOT,LDH and CPK,are higher than those in control,is in direct proportion to the H2O2 concentration and the incubation time.

Abstract:By Sepharose 4B-thyroglobulin affinity column chromatography a lectin was purified from Bellamya purificata. It showed a single band on alkaline PAGE. This lectin (BPL) agglutinated red blood cells of rabbit,duck and pig,but not of human blood types or formalin and/or glutaraldehyde-fixed rabbit erythrocytes. Its agglutination activity could be inhibited by 1.0mol/L lactose or galactose as well as 60g/L thyroglobulin, but not by alkaline borate buffer.BPL was temperature-sensitive,while showed a broad optical pH range (pH4.56-9.96).

Abstract:The expression of transforming growth factor α(TGFα)in human gastrointestinal carcinoma by methods of molecular hybridization and cell FIT cell-cycle analysis was investigated.The results show that the levels of TGFα mRNA are higher in gastric carcinoma tissues,colonic carcinoma tissues and rectal carcinoma tissues than in the corresponding control tissues. After treating with 12-O-tetradecanoylphorbol-13-acetate(TPA,100μg/L,4h),the level of TGFαmRNA in gastric carcinoma cell line(MGc80-3)increases,and percentage of S phase cells is markedly increased after 6 hours. These results suggest that TGF α plays an important role in gastrointestinal carcinoma tissues and expression of TGF α mRNA can be regulated by TPA.
