Abstract:Integrin is one kind of cell adhesion molecules which is distributed widely at cell surface. The structure and function of integrin was introduced.Integrin is a noncovalent associated heterodimer composed of α and β membrane-spanning subunits.It is involved in the adhesion between cell and cell,cell and extracelluar matrix.The signal transduction outside in was mediated by tyrosine kinase and protein kinase C. The alteration of integrin expression quality and quantity could be occured during carcinogenesis.

Abstract:Genomic subtraction is an efficient and economical method for isolating sequences pressent in one genomic DNA population(tester)but absent in another(driver).It proved widely applicable to detecting genomic abnormalities in cancer,probes for polymorphic loci and acquisition of new sequences in infectious diseases of viral organism. The fundation and development of genomic subtraction,experimental strategy,applications, problems in present and promise in future are described.

Abstract:Methane monooxygenase(MMO),an important enzyme in metabolism of methane in methanotrophs, catalyzes the hydroxylation of alkanes and the epoxidation of alkenes. In addition,MMO can biodegrade haloakanes and haloalkenes. Being one of the proteins containing binuclear iron centers,MMO can activate oxygen for insertion into a C-H bond.The elucidation of structures of active centers of MMO is significant for methane utilization and design of chemical catalysts. This article will concentrate on progress on above researches.

Abstract:General initiation factors SL1,TFⅡD and TFⅢB required for initiation of transcription in the three kinds of classic genes in eukaryotic cells respectively are multiprotein complexes that consist of a universal subunit termed TATA-box binding protein (TBP) and a set of TBP associated factors (TAFs).The C terminal domain of TBP, which is highly conserved in a wide variety of species, interacts directly with TATA element or binds to DNA through TAFs. TAFs are polymerase - specific and promoter-specific. In transcriptional initiation,some of the TAFs are expected to function as adoptors for TBP-DNA interaction, some of them may serve as bridging proteins that allow the other TAFs interact with TBP in the assembly of transcription initiation complex,and some as coactivators may mediate transcription initiation activated by sequence-specific activators.

Abstract:Synapsin Ⅰ is a specific phospho-protein of neuron,it is associated with the cytoplasmic surface of small synapsic vesicles. It is highly conservative in evolution and its gene locates in X-chromosome. By means of null mutation of synapsin Ⅰ gene in mice and investigation of brain slices electrophy-siologically,the mutation is generated by homologous recombination. And it is found that synapsin Ⅰ plays a vital role in physiological events such as neurotransmitter release and synaptic plasticity,etc.

Abstract:Various kinds of immunotoxin have been prepared with genetic engineering method.The most succcessful one among them is interleukin-2(IL-2) immunotoxin,which consists of IL-2 protein fused with pseudomonas exotoxin or diphtheria exotoxin. The IL-2 moiety in this type of immunotoxin acts as a targeting ingredient,so that to kill the target cells positive for IL-2 receptors. The IL-2 immunotoxin is being applied to treat transplantation rejections,autoimmune diseases and T cell lymphomas , etc. So far,a promising therapeutic efficiency has been achieved during the Ⅰ/Ⅱ phase clinical trials.

Abstract:Chinese hIFNcDNA deleted signal sequence was subcloned into downstream the PRPL promoter of the expression plasmid pBV220 by using of DNA recombinant technique.This recombinant plasmid can express hIFN-γ stably with high efficiency(up to 44.4% of the total bacterial soluble protein) in E. coli DH5α through thermal induction.The bioactivity assay showed that 0.45×107~2.34×107U rhIFN-γ were expressed in every liter of bacteria culture after preliminary purification and refolding procedures.

Abstract:Hypoxia and post-hypoxia reinfusion usually lead to a series of complicated physiological and biochemical changes.Through the model of mouse repetitive hypoxia ,the content changes of gangliosides and monoamine neurotransmitters were observed in the brain and it was found that:with the increment of the anoxic times:①Gangliosides(calculated as NANA)was continuously falling(P<0.01),and the falling of relative percentages of GM1 and GD1b of which were magnificent (GM1:P<0.05,GD1b:P<0.01);②Among monoamine neurotransmitters,the level of NE and DOPA descended and that of DA and HIAA increased (P<0.01).The results point out:after repetitive hypoxia,the cell membrane in centrical nerve tissues is injured in some degree and thus may affect the whole process of synthesizing,releasing,reabsorbing and storing of monoamines. There is association relation between changes of gangliosides and monoamines when hypoxia occurs.

Abstract:Anti-leukemic T-cell immunoliposomes entrapping cytotoxin from Chinese Cobra(CT)were prepared using dioleoylphosphatidylethanolamine (DOPE ) and monoclonal antibody(McAb)against human pan-T lymphocyte Wu71.The immunoliposomes is pH-sensitive,it is stable in pH7.0~8.0 but easy to release its contents in the slightly acidic environment of target cell plasma,below pH6.0.Treatment of the human T- leukemic cell line CEM with these immunoliposomes and using scanning electron microscopy to observe its dynamic killing action. The result shows that it was highly targetsensitive to the target cells while little cytotoxicity to the human non-target cells.

Abstract:The experimental results show that chemotaxis and adherence of polymorphonuclear neutrophil(PMN)are promoted obviously by exogenous phospholipase A2 and calcium ionophore(A23187). Furthermore,for the PLA2 inhibitor p-bromophenacyl bromide(PBPB) and the PLA2 antibody, the inhibitory effect on PLA2-induced PMN chemotaxis and adherence is concentration dependent but they have no effect on A23187-induced PMN chemotaxis and adherence. This result indicates that PLA2 induced chemotaxis and adherence of PMN may be inhibited by direct interaction between PLA2 and PBPB or PLA2 antibody. A23187 induced PMN chemotaxis and adherence is different from that induced by PLA2.

Abstract:Using site-directed mutagenesis method,six human interleukin-2 (IL-2) muteins with markedly reduced bioactivities were obtained,among them two muteins 15Val-IL-2 and 126Asp-IL-2 could decrease the biological effect of wild-type IL-2 in a specific concentration range. In assays of competition inhibition to high-affinity IL-2 receptor(IL-2R),15Val-IL-2 and 126Asp-IL-2 exhibited significant competitive abilities once again. These results showed that 15Val-IL-2 and 126Asp-IL-2 could partially antagonize the function of wild-type IL-2. Taking the analysis of IL-2 secondary structure and the knowledge of interaction between IL-2 and IL-2R into consideration,it may be concluded that the partial antagonism of 15Val-IL-2 and 126Asp-IL-2 lies in that substitution slightly disturb the tertiary binding microenviroment of IL-2 and IL-2R βγ subunits,thus interfering the binding of relevant IL-2 residues with IL-2R βγ subunits which yet is insufficient to totally disrupt such binding.

Abstract:A microcomputer software package for stereoscopic vision test named SVT was designed based on the principle of binocular disparity,Three kinds of patterns wete produced on color screen of PC computer:The first one was iso-disparity static random-dot stereogram(RDS)which contained eight kinds of patterns(such as circle,cross and on),six levels of disparity were:1'、3'、5'、10'、20'、30'.The second one was the pattern which disparity was different in different parts,such as bowl,ball and so on. And the last one was pattern with different frequencies of sinusoidal variation in disparity (included RDS and line-stereogram).A set of static RDS was used in testing stereoscopic vision for normal and abnormal vision children. It showed that the SVT software package can be used in amblyopia and strabismus screening clinically.

Abstract:Testosterone is prehormone of estradiol and most of it will converted to estradiol in ovary. The production of testosterone was regulated directly by luteinizing hormone and 17α-hydroxylase which is the rate-limiting enzyme in the production of testosterone.So 17α-hydroxylase and LHR gene expression reflec the response to LH of ovarian cells. In order to investigate the ovarian functions, the 17α-hydroxylase and LHR gene expression was detected simultaneously by using ribonuclease protection assay (RPA) and the sensitivity of RPA is about 8 times more compared with Northern blot.

Abstract:A microsome preparation procedure using polyethylene glycol(PEG )is reported. The liver homogenate of rats is aggregated by polyethylene glycol-6000,and centrifuged at high speed two times.The obtained pellet is the microsomal fraction. Compared with the ultracentrifugation method,this procedure of using PEG to prepare microsomes greatly eliminated the need for an ultracentrifuge and reduces the time necessary for isolation of this fraction. It is simple and feasible.

Abstract:α- and β-luffins were easily and quickly isolated and purified from seeds of Luffa cylindrica by using an improved procedure that involved ammonium suifate preciptation, ion exchange chromatography on S Sepharose Fast Flow and gel filtration on Sephadex G-75.α-and β-luffins were basic proteins with isoelectric points of about 10, with molecular weight of 28 000 and 29 000, respectively, as judged by SDS-PAGE. Inhibitory activities of α-luffin and β-luffin on cell-free protein synthesis were stronger than that of any known type-1 RIPs,with ID50 10μg/L and 50μg/L, respectively.

Abstract:Laboratory investigations for one step purification of recombinant human IL-4 (rhIL-4)with cation ion-exchange chromatography are presented.The method is easy to operate and gives no dilution effect on the sample.The purified rhIL-4 is of high purity (≥95%),high biological activity(≥10 MU/mg) and low toxicity(ET≤0.5 ZU/L),which meets the criteria of cytokines for the further experiemental research.Furthermore,purity of rhIL-4 can be increased as high as 98% with higher biological activity when two steps of purification were employed by using combination of ion exchange and gel-filtration chromatographies.

Abstract:Exon 5~8 of p53 gene in 10 cases of non small cell lung cancer(NSCLC)tissues and normal lung tissues were examined by nonisotopic polymerase chain reaction-single strain conformation polymorphism(PCR-SS-CP).It showed that there was point mutation in 4 cases of cancer tissues. The mutant points were 1 in exon 5~6,1 in exon 7,2 in exon 8 respectively. Otherwise,no point mutation was observed in normal lung tissues. Mutant p53 gene determined by PCR-SSCP was sequenced and found that codon ACA 280 instead of AGA 280,that is, cysteine instead of serine.
